چکیده

The genus Borrelia was known more than a century ago following the discovery of Borrelia recurrentis as the causative agent of epidemic relapsing fever. Interest in Borrelia increased dramatically after the recent discovery of Lyme disease and the observation that it is caused by a novel member of the genus. Borrelia are spirochaetes, organisms that comprise a separate phylum in the kingdom of eubacteria (Woese, 1987). They differ from other bacteria in having a helical shape with multiple waves and endoflagella which are located between the inner and outer membrane. In this review, we concentrate on the molecular biology of the causative agents of Lyme disease and North American relapsing fever (Borrelia btlrgdorferi senstl lato and Borrelia hermsii, respectively) because they have received the most research attention. To contain the review within reasonable limits we have not attempted to cover the taxonomy of the Borrelia or the widespread literature on antigenic variation of B. hermsii. Interested readers are referred to recent reviews on these topics by Barbour (1990) and Wilske e t al. (1992).

Evolution of the linear DNA replicons of the Borrelia spirochetes.

Members of the spirochete genus Borrelia carry numerous linear DNA replicons with covalently closed hairpin telomeres. The genome of one member of this genus, B. burgdorferi B31, has now been completely characterized and contains a linear chromosome, twelve linear plasmids and nine circular extra-chromosomal elements. The phylogenetic position of the Borrelia spirochetes strongly suggests that ...

Telomere resolution in the Lyme disease spirochete.

The genus Borrelia includes the causative agents of Lyme disease and relapsing fever. An unusual feature of these bacteria is a genome that includes linear DNA molecules with covalently closed hairpin ends referred to as telomeres. We have investigated the mechanism by which the hairpin telomeres are processed during replication. A synthetic 140 bp sequence having the predicted structure of a r...

The telomere resolvase of the Lyme disease spirochete, Borrelia burgdorferi, promotes DNA single-strand annealing and strand exchange

Spirochetes of the genus Borrelia include the tick-transmitted causative agents of Lyme disease and relapsing fever. They possess unusual genomes composed mainly of linear replicons terminated by closed DNA hairpin telomeres. Hairpin telomeres present an uninterrupted DNA chain to the replication machinery overcoming the 'end-replication problem' for the linear replicons. Hairpin telomeres are ...

The Borrelia burgdorferi telomere resolvase, ResT, possesses ATP-dependent DNA unwinding activity

Spirochetes of the genus Borrelia possess unusual genomes harboring multiple linear and circular replicons. The linear replicons are terminated by covalently closed hairpin (hp) telomeres. Hairpin telomeres are formed from replicated intermediates by the telomere resolvase, ResT, in a phosphoryl transfer reaction with mechanistic similarities to those promoted by type 1B topoisomerases and tyro...

Linear plasmids of Borrelia burgdorferi have a telomeric structure and sequence similar to those of a eukaryotic virus.

Spirochetes of the genus Borrelia have double-stranded linear plasmids with covalently closed ends. The physical nature of the terminal connections was determined for the 16-kb linear plasmid of the B31 strain of the Lyme disease agent Borrelia burgdorferi. Native telomeric fragments representing the left and right ends of this plasmid were isolated and subjected to Maxam-Gilbert sequence analy...
